First of all, copy all the data you need elsewhere.

On Windows, open disk management and delete all partitions of the stick. Watch that you delete the stick and not those from your hard drives. Therefore double check the disk size!

After deleting, you’ll create a single partition. FAT if you want to put the stick in other devices like TV, Blu-Ray Player etc. because they can often only FAT.

Efficient and if files should have more than 4GB, then take NTFS. Then the stick does not work anymore with any “not PC” device.
